应用 RT- PCR技术对 1株分离于地方免疫鸡群中暴发的传染性法氏囊病病例的传染性法氏囊病病毒 ( infec-tious bursal disease virus,IBDV) HN0 2 6株的 VP2基因进行了克隆与序列分析 ,并与相应毒株的 VP2高变区核苷酸及氨基酸序列进行了比较研究。结果表明 ,HN0 2 6株的核苷酸和氨基酸序列与变异株 Var- A的同源性最高 ,分别达97.3%和 97.6 % ;其次为超强毒株 UK6 6 1 ,分别为 95 .7%和 95 .2 % ;而和弱毒株 PBG98的同源率仅有 92 .8%和90 .3%。其中 ,在第一、二亲水区 ,HN0 2 6株均有 1个氨基酸发生了变化 ,即第 2 2 2位转变为 E、第 31 8位转变为 D。更重要的是 ,其 2 4 9位和 2 5 4位上的氨基酸分别为 K和 S,这些均为变异株的特性。另外 ,HN0 2 6株的七肽区保持SWSASGS不变 ,且第 2 79位和 2 84位氨基酸分别为 D和 A,又完全具备强毒株的特性。因此 ,初步确定所分离的HN0 2 6为有较强致病力的变异株 IBDV。 相似文献

Background
In veterinary medicine, congenital methemoglobinemia associated with nicotinamide adenine dinucleotide (NADH)‐cytochrome b5 reductase (b5R) deficiency is rare. It has been reported in several breeds of dogs, but little information is available about its etiology.Objectives
To analyze the NADH‐cytochrome b5 reductase gene, CYB5R3, in a Pomeranian dog family with methemoglobinemia suspected to be caused by congenital b5R deficiency.Animals
Three Pomeranian dogs from a family with methemoglobinemia were analyzed. Five healthy beagles and 5 nonrelated Pomeranian dogs without methemoglobinemia were used as controls.Methods
Methemoglobin concentration, b5R activity, and reduced glutathione (GSH) concentration were measured, and a turbidity index was used to evaluate Heinz body formation. The CYB5R3 genes of the affected dog and healthy dogs were analyzed by direct sequencing.Results
Methemoglobin concentrations in erythrocytes of the affected dogs were remarkably higher than those of the control dogs. The b5R activity of the affected dogs was notably lower than that of the control dogs. DNA sequencing indicated that this Pomeranian family carried a CYB5R3 gene missense variant (ATC→CTC at codon 194) that resulted in the replacement of isoleucine (Ile) by leucine (Leu).Conclusions and Clinical Importance
This dog family had familial congenital methemoglobinemia caused by b5R deficiency, which resulted from a nonsynonymous variant in the CYB5R3 gene. This variation (c.580A>C) led to an amino acid substitution (p.Ile194Leu), and Ile194 was located in the proximal region of the NADH‐binding motif. 初步探讨了在‘越心’草莓(Fragaria × ananassa‘Yuexin’)茎尖离体培养再生植株中发现的稳定变异突变体着色差异的分子机理。分析结果显示,突变体与‘越心’在植株形态、始花期、始果期、始熟期、单果质量、平均株产、果形、风味等方面均无明显差异;而突变体外果皮色泽较淡,L*值较高,a*值较低,H色度角值较大,颜色指数CIRG显著小,且果肉不着色(‘越心’果肉红色)。突变体果实总花青苷含量为‘越心’的13.2%,其中以天竺葵素–3–O–葡萄糖苷(Pg3G)含量差异最大,突变体Pg3G含量仅为‘越心’的12.9%。花青苷合成途径结构基因FaF3H、FaCHS、FaDFR、FaANS、FaCHI、Fa3GT的表达水平在突变体中显著降低,这可能是突变体成熟果实花青苷水平显著低于‘越心’的直接原因。FaMYB10和FaMYB1在突变体中的表达水平显著下降,且FaMYB10在几个主成分中的特征向量绝对值均显著较高,进一步证实了MYB10是草莓成熟过程中花青苷合成的类黄酮或苯丙烷类化合物途径主要调节因子之一。提取叶片基因组DNA进行基于KASP标记的基因分型结果显示,74对KASP引物在‘越心’草莓及其突变体中的基因分型不存在差异。本结果初步证实突变导致参与调控花青苷合成途径的关键基因表达发生显著下调从而使花青苷积累显著减少,果实着色变淡。 相似文献

针对晋阳345和晋麦73两个定型品种存在抗寒性较差的缺点,对其幼胚愈伤组织进行了4种处理:1、愈伤组织在MD培养基上继代2~3次;2、愈伤组织在紫外灯下约10cm照射2h;3、愈伤组织在低温2℃和常温25℃下交替培养30d;4、愈伤组织在紫外灯下照射2h后,再在低温2℃和常温25℃下交替培养30d。结果表明,4个处理后代R2主要农艺性状的变异率差异显著,且紫外灯照射幼胚愈伤组织的变异率比低温2℃和常温25℃交替处理幼胚愈伤组织高,其后代各种农艺性状的变异范围较大,变异率较高。但后者有利于改良抗寒性较差的定型小麦品种。 相似文献

To search natural compounds having inhibitory effect on bacterial growth is important,
particularly in view of growing multidrug resistant (MDR) strains of bacterial pathogens.
Like other bacterial pathogens, MDR Vibrio cholerae, the causative agent
of diarrheal disease cholera, is becoming a great concern. As an approach of searching new
antimicrobial agents, here, we show that anethole, a well-studied natural component of
sweet fennel and star anise seeds, could potentially inhibit the growth of MDR O1 El Tor
biotype, the ongoing 7th cholera pandemic variant strains of toxigenic V.
cholerae. The minimum inhibitory concentration (MIC) of anethole against
diverse O1 El Tor biotype strains is evaluated as 200
µg/ml. Moreover, the effect of anethole is
bactericidal and exerts rapid-killing action on V.
cholerae cells. This study is the first report which demonstrates
that anethole, purified from natural compound, is a potent inhibitor of growth of
toxigenic V. cholerae. Our data suggest that anethole could be a
potential antimicrobial drug candidate, particularly against MDR V.

Adverse events can occur after rabies post‐exposure prophylaxis (PEP), and linkage to causality is often difficult to determine. We report a case of recurrent temporary paralysis that began immediately after the initiation of rabies PEP in a man exposed to a bat. The recurrent temporary paralysis first occurred in the patient after his initial dose and then again after day 3 of his rabies PEP. The PEP was terminated prior to a serologic response. 在已构建致仔猪水肿病大肠杆菌S451521菌株的2个毒素敲除菌株(F1、F2)基础上,进一步测定了其毒力及在仔猪肠道内的繁殖能力。选择30日龄的ICR小鼠并随机分成4组,A、B、C试验组分别灌胃接种不同浓度的野生菌株S451521及其毒素敲除菌株F1、F2,D组以灭菌生理盐水作阴性对照;结果表明减毒菌株F1和F2毒力均比野生菌株弱,其中F1株减弱了60.73%、F2株减弱了84.38%。选择24日龄未经仔猪水肿病疫苗免疫过的仔猪共14头,并随机分为2组;第1组口服接种减毒菌株F2,第2组以灭菌生理盐水作阴性对照,定期采集仔猪粪样并进行细菌监测;结果显示,仔猪排出减毒菌株F2的数量随时间不断增多,在接种后28d可达到每克粪样含目的菌约70 000cfu。这说明减毒菌株F2能够在仔猪体内定植并大量繁殖。 相似文献

Two variants of Loma salmonae occur in net-pen reared chinook salmon, Oncorhynchus tshawytscha. The typical variant (OA) has a host specificity for salmonids of the genus Oncorhynchus whereas the atypical variant (SV) has a host specificity for brook trout, Salvelinus fontinalis, and in this study, the ultrastructure of the two are compared. In fish at 8 weeks post-exposure xenomas of the SV variant have a very high proportion of mature spores compared with other developmental stages, while in xenomas of the OA variant there are fewer spores and many other developmental stages. Spores of the SV variant had up to 20 turns of their polar tube whereas those of the OA variant only had 17. Furthermore, the spores of the SV variant were significantly larger than those of the OA variant. The sporophorous vesicle for both variants appears to form around a sporogonial plasmodia, which results in many spores developing within the vesicle. 相似文献
